Eyeshield 21 cuenta la historia de un chico llamado Sena Kobayakawa, un joven que no destaca mucho, al contrario. Muchas veces es maltratado por sus compañeros, pero tiene una gran habilidad en sus piernas, lo que le permite correr a gran velocidad. Después de presentar su examen de admisión, Sena entra a la preparatoria Deimon (aunque luego se sabe que por falta de alumnos todos entraron a pesar de aprovar o suspender), junto con su amiga de la infancia Mamori Anezaki, es en la preparatoria Deimon donde comienza su aventura en el club de fútbol-americano, donde entra accidentalmente para luego convertirse en eyeshield 21, el hombre con la velocidad de la luz en sus piernas y donde luchara para conseguir el sueño de su equipo: llegar al Chrismas Bowl el torneo más importante a nivel instituto de Japón.
