Als es durch Krieg und Seuchen schlecht um seinen Einflussbereich steht, schwört Daigo Kagemitsu, Burgherr und Vasall des Feudalherren der Provinz Kaga, dem Buddhismus ab und betet stattdessen zwölf Dämonen an, ihm Macht zu verleihen, was es auch kosten mag. Bei der Geburt seines Sohnes stellt sich heraus, welchen Preis er für sein Bitten bezahlen musste, denn dieser kommt verstümmelt, ohne Gliedmaßen, Haut oder Augen zur Welt. Berauscht davon, die Dämonen nun auf seiner Seite zu wissen, lässt Daigo den Säugling in einen Fluss werfen.
Sechzehn Jahre später trifft ein elternloses Kind namens Dororo, das sich mit verschiedenen Gaunereien über Wasser hält, bei seiner Flucht vor ein paar Ganoven auf einen jungen Mann in zerlumpten Kleidern, der eine Maske und Prothesen anstelle seiner Gliedmaßen trägt. Als unvermittelt ein in Schlamm gehüllter Dämon aus einem nahe gelegenen Fluss aufsteigt und Dororos Verfolger verschlingt, besiegt der Maskierte das Wesen trotz seines Handicaps scheinbar mit Leichtigkeit. Begeistert von den Fähigkeiten des Unbekannten beschließt Dororo, diesen auf seiner Reise zu begleiten …
