Als „Stern am Hof der Medici“ und „Freigeist“ wird Isabella de Medici in ihrer Zeit gerühmt. Doch in einer von Männern beherrschten Welt sorgt ihre Ausnahmestellung auch für Neid und Missgunst – sogar in der eigenen Familie. Nach dem Tod ihrer Mutter steigt Isabella, die geliebte Tochter Cosimos I., zur „First Lady“ von Florenz auf. Die strahlend schöne, charmante und kluge Isabella wird die engste Vertraute ihres Vaters, erfolgreich leitet sie den Familienbetrieb, glänzend repräsentiert sie die Dynastie. Neiderfüllt muss der Thronfolger, Isabellas Bruder Francesco, dem Aufstieg seiner Schwester zusehen. Alles ändert sich, als Cosimo I. 1574 stirbt. Von nun an setzt der neue Herrscher Francesco in der Nachfolge seines Vaters seine Macht ohne Skrupel ein. Er sorgt dafür, dass Isabella das Erbe Cosimos I. nicht erhält. Er fordert sie auf, zu ihrem Mann zurückzukehren, und er macht sie für das schlechte Benehmen ihrer Cousine Dianora verantwortlich. Doch Isabella wehrt sich. Sie ist bereit, für ihr Recht und ihre Freiheit zu kämpfen. Als eine Verschwörung gegen Francesco aufgedeckt wird und Dianora einem der Beschuldigten zur Flucht verhilft, wird die Situation für Isabella und ihre lebenslustige Cousine schwierig. Der erste Teil des Doku-Dramas begibt sich auf die Suche nach den sterblichen Überresten Isabella de Medicis und rekonstruiert ihre dramatische Lebensgeschichte. Kann nach über 400 Jahren der verschwundene Leichnam Isabellas endlich identifiziert werden? Und warum versuchte man damals, die Erinnerung an den „Stern von Florenz“ für immer zu tilgen? Die Wissenschaft hat es sich zum Ziel gesetzt, diese Fragen zu beantworten.
