被一只有辐射的蜘蛛咬了一口后，年轻的彼得·帕克发现自己拥有了蜘蛛般的超能力。他因此被自负蒙蔽，希望能利用超能力获得名气和财富，却忽视了别人的需求，最终因拒绝帮警察抓几个逃跑的罪犯而间接导致了他的叔叔本死去。被这巨大的挫折所打击，彼得决心用自己的超能力去打击罪恶，并变成了超级英雄蜘蛛侠。在对付超级恶人们的同时，彼得还要平衡自己的个人生活，包括和女朋友玛丽·简的感情，以及《号角日报》的摄影和编辑工作。作为一名编辑，他坚信蜘蛛侠是个罪犯，必须被打倒。
