近藤勇が新選組の実権を握ってから池田屋襲撃に至るまで、新選組の最も華やかだった頃に焦点をあてる。新選組を題材にした作品がこれまでに数多く存在する中で、今作は理屈抜きに楽しめる時代劇としての“新選組”の物語を徹底的に追求。幕末という激動の時代の只中で真剣に生きた男たちを真正面から描いている。また、新選組のほかに乱世の中で空しく死んでいった志士・浪士たちの家族にも着目している点も見どころのひとつ。新選組は、京の治安を守るために会津藩の預かり組織として設立された。しかし、初代局長・芹沢鴨（亀石征一郎）はその権力を悪用して悪行・乱行を繰り返し、新選組は京の人々から忌み嫌われる存在となっていた。この事態を憂いた近藤勇（里見浩太朗）は、土方歳三（地井武男）や沖田総司（野村宏伸）らとともに実力行使で芹沢を退ける。そして近藤は、自ら局長となって厳しい隊規で部下を律した。その結果、新選組に対する市民の信頼は回復していった。その頃、新選組擁護派の要人たちが次々と暗殺された。会津藩主・松平容保（田中健）は近藤を呼び、浪士たちの取り締まりを一層強化するよう命じる。それから、新選組による厳しい浪士狩りが展開される…。
