Die beiden Mädchen gehen auf die zweite Klasse der Verone Mittelschule, auf der Nagisa im Lacrosse-Team spielt und Honoka im Wissenschaftsclub tätig ist. Mepple und Mipple, zwei Flüchtlinge aus dem Garten des Lichts, verhelfen den zwei erstaunten Mädchen zu Superkräften, welche sie auch sogleich benötigen, da ihnen ein Gegenspieler aus der Dusk Zone entgegentritt. Die Dusk Zone mit ihrem Herrscher Dark King ist hinter den sieben Prismasteinen her, mit deren Hilfe sie den Garten des Lichts und auch andere Welten beherrschen wollen. Nagisa verwandelt sich in Cure Black, Honoka sich in Cure White. Zusammen sind sie Pretty Cure. Als solche versuchen sie, die fünf Prismasteine, die sich schon im Besitz des Dark King befinden, zu bekommen, um den Garten des Lichts wieder herzustellen.
